Molecular cloning of WIF1 and HMGA2 reveals ear‐preferential expression while uncovering a missense mutation associated with porcine ear size in WIF1

Zusammenfassung: | <jats:title>Summary</jats:title><jats:p>Considerable diversity exists in porcine ear size, which is an important morphological feature of pig breeds. Previously, we localized four crucial candidate genes—<jats:italic>high mobility group <jats:styled-content style="fixed-case">AT</jats:styled-content>‐hook 2</jats:italic> (<jats:italic><jats:styled-content style="fixed-case">HMGA</jats:styled-content>2</jats:italic>), <jats:italic><jats:styled-content style="fixed-case">LEM</jats:styled-content> domain‐containing 3</jats:italic> (<jats:italic><jats:styled-content style="fixed-case">LEMD</jats:styled-content>3</jats:italic>), <jats:italic>methionine sulfoxide reductase B3</jats:italic> (<jats:italic><jats:styled-content style="fixed-case">MSRB</jats:styled-content>3</jats:italic>) and <jats:italic>Wnt inhibitory factor 1</jats:italic> (<jats:italic><jats:styled-content style="fixed-case">WIF</jats:styled-content>1</jats:italic>)—on <jats:italic>Sus Scrofa</jats:italic> chromosome 5 affecting porcine ear size, then cloned <jats:italic><jats:styled-content style="fixed-case">LEMD</jats:styled-content>3</jats:italic> and <jats:italic><jats:styled-content style="fixed-case">MSBR</jats:styled-content>3</jats:italic>. In this study, we performed rapid amplification of <jats:styled-content style="fixed-case">cDNA</jats:styled-content> ends to obtain full‐length <jats:styled-content style="fixed-case">cDNA</jats:styled-content> sequences of 2338‐bp <jats:italic><jats:styled-content style="fixed-case">WIF</jats:styled-content>1</jats:italic> and 2998‐bp <jats:italic><jats:styled-content style="fixed-case">HMGA</jats:styled-content>2</jats:italic>. Using quantitative real‐time <jats:styled-content style="fixed-case">PCR</jats:styled-content>, we revealed that <jats:italic><jats:styled-content style="fixed-case">WIF</jats:styled-content>1</jats:italic> expression was highest in ear cartilage of 60‐day‐old pigs and that this is therefore a better candidate gene for ear size than <jats:italic><jats:styled-content style="fixed-case">HMGA</jats:styled-content>2</jats:italic>. We further screened coding sequence variants in both genes and identified only one missense mutation (<jats:italic><jats:styled-content style="fixed-case">WIF</jats:styled-content>1:</jats:italic>c.1167C>G) in a conserved epidermal growth factor‐like domain from the mammalian <jats:styled-content style="fixed-case">WIF</jats:styled-content>1 protein. The protein‐altering mutation was significantly associated with ear size across the Large White × Minzhu hybrid and Beijing Black pig populations. When <jats:italic><jats:styled-content style="fixed-case">WIF</jats:styled-content>1:</jats:italic>c.1167C>G was included as fixed effect in the model to re‐run a genome‐wide association study in the Large White × Minzhu intercross population the <jats:italic>P</jats:italic>‐value of the peak <jats:styled-content style="fixed-case">SNP</jats:styled-content> on <jats:styled-content style="fixed-case">SSC</jats:styled-content>5 from re‐running the genome‐wide association study dropped from 2.45E‐12 to 7.33E‐05. Taken together, the <jats:italic><jats:styled-content style="fixed-case">WIF</jats:styled-content>1:</jats:italic>c.1167C>G could be an important mutation associated with ear size. Our findings provide helpful information for further studies of the molecular mechanisms controlling porcine ear size.</jats:p> |
